Upload failed error. Please help!

Hi All,
I've connected my Mega 2560 R3 which I've just got to my PC and powered it through a 12v PSU and connected it to the USB port. I've run the config utility and downloaded a firmware update which I then tried to apply. After a few minutes I got an error saying Upload Failed!! What do I do?

You need to select the Arduino Mega option from the Firmware drop down on the right.
